چکیده
Rhododendron decorum is a woody species with high ornamental and medical value. Herein, we introduce novel in vitro regeneration method for R. decorum. We used flower buds to develop an efficient rapid plant protocol. Sterile of 2 cm size were as explants study the effects culture medium growth regulators on callus induction adventitious shoot differentiation, proliferation, rooting. According results, optimal combination was WPM + 1 mg/L TDZ 0.2 NAA, its rate reached 95.08%. The differentiation from 0.5 0.1 91.32%. proliferation ZT which 95.32% coefficient 9.45. rooting shoots NAA IBA, 86.90%. survival rates rooted regenerated plantlets exceeded 90% after acclimatization transplantation. This system has advantages being simple highly efficient, it causes little damage mother plants, laying foundation plantlet propagation, genetic transformation, new-variety breeding

Ice nucleation activity in various tissues of Rhododendron flower buds: their relevance to extraorgan freezing
Wintering flower buds of cold hardy Rhododendron japonicum cooled slowly to subfreezing temperatures are known to undergo extraorgan freezing, whose mechanisms remain obscure. We revisited this material to demonstrate why bud scales freeze first in spite of their lower water content, why florets remain deeply supercooled and how seasonal adaptive responses occur in regard to extraorgan freezing...
متن کاملAn efficient regeneration and rapid micropropagation protocol for Almond using dormant axillary buds as explants.
An efficient in vitro protocol was standardized for Almond (Prunus dulcis) propagation using dormant axillary buds as explants. Explants were cultured on Murashige and Skoog (MS) and woody plant medium (WPM) supplemented with different concentration/combination(s) of phytohormones. MS basal medium showed lowest shoot induction and took longest duration for shoot initiation. Multiple shoots were...
متن کاملLive confocal imaging of Arabidopsis flower buds.
Recent advances in confocal microscopy, coupled with the development of numerous fluorescent reporters, provide us with a powerful tool to study the development of plants. Live confocal imaging has been used extensively to further our understanding of the mechanisms underlying the formation of roots, shoots and leaves. However, it has not been widely applied to flowers, partly because of specif...
متن کاملProtecting Stone Fruit Flower Buds from Winter Freeze Damage
Flower buds of peach, apricot, and sweet cherry are killed by low temperatures during winter and spring. Frost protection measures used commonly in the spring are applied to freeze protection during the winter in the Yakima Valley of Washington. Critical temperatures change rapidly during winter. To succeed, winter freeze protection requires adequate inversions, equipment that operates at tempe...
متن کاملSupercooling in overwintering azalea flower buds.
Differential thermal analysis and nuclear magnetic resonance spectroscopy experiments on whole flower buds and excised floral primordia of azalea (Rhododendron kosterianum, Schneid.) proved that supercooling is the mode of freezing resistance (avoidance) of azalea flower primordia. Increase in the linewidth of nuclear magnetic resonance spectra for water upon thawing supports the view that inju...
